Inducing Apoptosis in Cancer by Targeting NF-κB and C/EBP
NF-!B is a protein that inhibits cell death in a large variety of malignancies. We have identified a novel pathway for activation of NF-!B regulated genes involving interaction of the p50 subunit of NF-!B with C/EBP proteins. Targeting this pathway may prove effective in cancers that lack the classical NF-!B activation pathway and in cancers that become resistant to drugs that target the classical pathway.
The goals of their present proposal are to identify human cancers that depend on C/EBP interaction with NF-!B p50 for survival, focusing initial efforts on breast cancer, and to map the contact points between the two proteins to allow computer aided design of novel therapeutics.
